Application of total organic carbon determination method in pharmaceutical water
organic substances in pharmaceutical water generally come from water sources, water systems (including purification, storage and transportation systems) and the growth of biofilm in water systems. Total organic carbon reflects and represents the amount of organic substances in pharmaceutical water
the key supporting components are similar products of international and domestic famous brands. Sucrose is usually used as an organic substance that is theoretically easy to oxidize, and the standard solution is configured according to the inspection limit for testing. The corresponding response value is obtained on the total organic carbon tester. 1,4-benzoquinone is used as an organic substance that is theoretically difficult to oxidize, and the system applicability test is carried out with its preparation solution to examine the oxidation capacity of the instrument
the principle of the total organic carbon determination method is to completely oxidize the organic matter molecules in the water into carbon dioxide (CO2), detect the amount of carbon dioxide generated, and then calculate the concentration of organic carbon in the effluent. There are two forms of carbon in pharmaceutical water, inorganic carbon and organic carbon, so there are usually two methods to determine total organic carbon: one is to subtract the measured inorganic carbon from the measured total carbon (inorganic carbon and organic carbon); The other is to remove inorganic carbon before the oxidation process
due to organic pollution and carbon dioxide, but this unrealistic absorption will affect the authenticity of the determination results. Therefore, attention should be paid to avoid pollution in all links of determination. The closed container shall be used for sampling, and the head space of the container shall be as small as possible. Test immediately after sampling. Organic residues must be strictly removed from the glassware used, and the final rinsing must be done with total organic carbon inspection water
1. General requirements for instruments
the instruments used should be calibrated, and the applicability of the instruments should be tested regularly with standard solution according to the specified method. The minimum carbon detection limit is required to be 0.05mg per liter or less
2. Total organic carbon inspection water
should be used per liter, so how to test the viscosity of the tape? This requires special instruments to test high-purity water with total organic carbon less than 0.10mg and conductivity less than 1.0us/cm (25 ℃). The water used for total organic carbon inspection and the water used for preparing the control solution and system suitability test solution should be contained in the same container
3. Reference solution
sucrose reference solution unless otherwise specified, take an appropriate amount of sucrose reference solution dried to constant weight at 105 ℃, accurately weigh it, add total organic carbon, dissolve it with water for examination, and dilute it to make a solution containing 1.19mg per liter (0.50mg per liter of carbon)
1,4 p-benzoquinone reference solution unless otherwise specified, take an appropriate amount of 1,4 p-benzoquinone reference solution, accurately weigh it, add total organic carbon inspection water to dissolve and dilute it to make a solution containing 0.75mg per liter (0.50mg per liter of carbon)
4. The system uses the pollutant content test of electroplating sewage
take the total organic carbon inspection water, inject the sucrose reference solution and 1,4 p-benzoquinone reference solution respectively, and record the total organic carbon response value of the instrument in turn. According to the following formula, the corresponding efficiency of the instrument should be
85% ~ 115%
[(rss-rw)/(rs-rw)] × 100
where RW is the response value of total organic carbon inspection water
rs is the response value of sucrose control solution
rss is the response value of 1,4 p-benzoquinone reference solution
5. Determination method
take an appropriate amount of pharmaceutical water for the test, test according to the specified method of the instrument, and record the response value Ru of the instrument. The response value of water for pharmaceutical use should be less than RS RW (0.50mg/l)
